报告内容简介:
The fungal kingdom is remarkable for the production of bioactive natural products commonly referred to as secondary metabolites (SMs). The study of fungal secondary metabolism has recently garnered significant interest, and the advent of fungal genomics has led to new insights into SM discovery and regulatory mechanisms. Much of this activity is driven by interests in drug discovery due to the biological properties of these metabolites. However, the treasure of SM is far underestimated and unexplored from the sequenced genomes because most of the biosynthetic genes are silent in the laboratory conditions. Here we describe key technological breakthroughs that together result in advances in mining the fungal SMs. These include: (1) we identified a series of SM restorers and novel metabolites by manipulating global regulator LaeA; (2) prenyltransferase-guided discovery of fungal indole alkaloid and (3) novel polyketide production in a heterologous expression via synthetic biological approaches. Taken together, our studies provide the efficient ways for finding new SM regulators and bioactive metabolites.
